OUR SCIENCE
Using radiation as
a chemical trigger.
PolyAdvant is developing therapeutic conjugates designed to respond to ionizing radiation and release active payload in the treatment field.
THE OPPORTUNITY
Spatial precision,
translated into chemistry.
Radiotherapy already delivers energy to a defined treatment region. Our platform explores how that same energy can initiate a chemical transformation in a therapeutic conjugate.
This approach differs from conventional chemoradiotherapy, where chemotherapy generally acts systemically and may sensitize tumors to radiation. Radiation-activated prodrugs are instead designed for externally triggered drug release.
CONCEPTUAL MECHANISM
Masked therapeutic
A targeting component carries a payload in a designed conjugate.
Radiotherapy
Ionizing radiation is applied to the planned treatment field.
Active payload
Responsive chemistry aims to enable local activation alongside radiation.
This diagram presents the intended platform concept and is not a representation of established clinical performance.
PLATFORM DESIGN
A modular approach to
radiation-responsive therapy.
The platform brings together targeting, linker chemistry and payload selection within a treatment strategy built around radiotherapy.
External control
Radiotherapy offers a clinically established, spatially planned source of activation energy.
Modular delivery
The platform can be explored across small-molecule conjugates and antibody–drug conjugates.
Combination by design
Drug activation and radiotherapy are considered together from candidate design through treatment scheduling.

Switching on prodrugs using radiotherapy
The study demonstrated proof of concept for gamma/X-ray-mediated activation of caged fluorescent probes and anticancer prodrugs at clinically relevant radiation doses.
